PATHOGENIC INTESTINAL VIRUS ECHO 7-KOR/95 ISOLATED FROM KOREAN PATIENTS, AND COMMON ANTIGEN AGAINST INTESTINAL VIRUS PREPARED THEREFROM

PURPOSE: A pathogenic intestinal virus Echo 7-Kor/95 isolated from Korean patients, a common antigen against the intestinal viruses, and DNA encoding the antigen are provided. The antigens are used in rapidly diagnosing diseases caused by the intestinal virus and in developing vaccines. CONSTITUTION: The pathogenic intestinal virus Echo 7-Kor/95 belongs to Picornaviridae sp. and induces abacterial cerebral meningitis, keratoconjunctivitis, and hand-foot-and-mouth disease. Echo 7-Kor/95 is isolated from the feces of Korean patients suffering from abacterial cerebral meningitis and identified. The RNA of Echo 7-Kor/95 is extracted, cDNA is then synthesized from the RNA, and cDNA is amplified by PCR. A cDNA library is constructed by inserting the cDNA into an appropriate vector. cDNA clones exhibiting positive reaction for serum of patients suffering from diseases, which are caused by intestinal viruses, are identified among the cDNA libraries by using an immune screening method. The base sequences of cDNA clones exhibiting positive reaction are analyzed and the coding region of a common antigen is then identified. The coding region is amplified by PCR and protein is expressed from the amplified DNA. The operation of the protein as a common antigen for intestinal viruses is tested.
