PROBLEM TO BE SOLVED: To shorten a time required in the examination of the concn. of a substance having a rotatory power such as sugar and protein contained in urine by a simple constitution and to detect the concn. of the substance highly reliably without being affected by noises or the like. SOLUTION: A magnetic field is applied to the light path from a light emitting means 11 to a light receiving means 12 by a magnetic field applying means 4. This applied magnetic field is changed over to a plurality of stages by a magnetic field switching means 16, and a modulation signal of a specific cycle is superposed on the magnetic field by a magnetic field modulating means 18. The output of the light receiving means 12 is sampled by a sampling means 14 and Fourier coefficients of two signal components equal to the modulation frequency due to the magnetic field modulating means and different in phase byπ/2 are calculated from the data row obtained by sampling by a frequency component calculating means 19 and the square sum of them is calculated to simply detect concn. with high reliability for a short time without being affected by noises or the like.
