The present invention relates to controlled modification of Src homology region 3 (SH3) domains by using random manipulation of the non-conserved region of SH3 RT-loop. Consequently, the invention concerns methods and materials for generating SH3 domains with engineered binding properties, and their use as tools in research, diagnostics, therapy and drug discovery.
