C121 DNA is successfully isolated from an ST2 cDNA library, which is a mouse myeloid stroma-origin cell line, by an expression cloning method with the use of a retrovirus. A protein encoded by this cDNA is a complete protein comprising a partial amino acid sequence of mouse ISF protein. It occurs as a soluble protein and a cell membrane-attached protein and both of these types show an activity of supporting the proliferation of S21 cells. Also, a human cDNA corresponding to mouse ISF gene is successfully isolated from a human kidney-origin cDNA library.
