摘要 The invention relates to a process for preparing urease from Citrullus vulgaris seeds, consisting in refluxing 200 parts by weight of water melon seed flour together with 1,000 parts by volume of a non-polar solvent for 3 hours, in order to eliminate fatty acids and release the enzyme which is extracted from the remaining cake with a phosphate buffer solution with pH=7.0, is purified in a chromatographic column with a slightly basic dextrane-based ion exchange resin, with the functional group diethylaminoethyl and with the replacing ion Cl- and is crystallized by precipitation with acetone, finally producing an enzyme having a purity degree of 320 U/mg of protein.
