PURPOSE: A vector, which comprises a polysaccharide decomposing enzyme gene as a selection marker, is provided. The using method thereof is easier than a vector comprising beta-galactosidase as a selection marker, is cost effective and is applied to a broad range of a host cell. CONSTITUTION: A desired vector, i.e., pCEL1 comprises a polysaccharide decomposing enzyme gene as a selection marker. A site of a restriction enzyme of the pCEL1 is as described in figure 1. The polysaccharide-decomposing enzyme that is introduced into a plasmid is selected one of cellulose, amylase and xylanase. The host is selected from E. coli, yeast and Lactobacillus. A method of selecting the transformant is characterized by comprising the steps of: introducing the target foreign gene into the pCEL1; transforming a host with the vector; smearing the transformed host on polysaccharide-containing medium and culturing then it; adding pigment reagent into the medium; and selecting finally the desired transformant by confirming if coloration occurs.
