A lipid bilayer vesicle comprising a).a membrane that has a positive net charge and 5 to 30% by weight, based on total lipids, of 1,3-dioleoyloxy-2-(6-carboxy-spermyl)-propyl-amide (DOSPER) and other lipids b).at least one viral hemagglutinin fusogenic peptide that causes the vesicles to be internalized by target cells through phagocytosis or endocytosis. c).A desired drug or nucleic acid material Also disclosed is a process for the manufacture of the above vesicle by a).dissolving the purified virus with a non-ionic detergent solution comprising C12E8 (octaethyleneglycol monododecylether) or n-octyl-oligooxyethylene, and obtaining by centrifugation, a supernatant fragment containing hemagglutinin b).preparing a solution containing : a buffer; a non-ionic detergent; part of the hemagglutinin supernatent, DOSPER (5-30% by weight of total lipids) and other lipids, and c).removing the detergent by dialysis or by treating the solution with microcarrier beads to form vesicles. Such vesicles are useful for carrying out non-infectious gene therapy.
