GUINEA PIG SKIN DISK, METHOD OF PREPARATION, AND METHOD OF ANTIBIOTIC MEASUREMENT

PURPOSE: A method for producing Guinea pig skin disc and a method for analyzing antimicrobial activity using the Guinea pig skin disc are provided, thereby the absorbability and antimicrobial activity of medicines on the skin can be simultaneously analyzed to inhibit the use of an excessive amount of medicines for the test. CONSTITUTION: The method for producing Guinea pig skin disc comprises the steps of: cutting hairs of Guinea pigs, peeling the belly skin from Guinea pigs, and dipping it in 200 to 500% of water at 24 to 40 hours; dipping it in a solution containing 200 to 500% of water, 2 to 5% of CaO/Ca(OH)2, 2 to 5% of sodium sulfate solution, and 1 to 5% of nonionic surfactant for 24 hours and washing it with water for 20 to 30 minutes; dipping it in 1 to 5% of calcium hydroxide solution for 2 to 3 days and washing it with water; dipping it in 200 to 500% of water containing 1.5% of ammonium sulfate for 1 hour and washing it with water; drying the Guinea pig skin on the plate; and punching the Guinea pig skin in an appropriate size. The method for analyzing antimicrobial activity comprises the steps of: sterilizing the Guinea pig skin disc with alcohol and dipping it in distilled water; soaking the disc in sample solution and washing it with water; placing the disc on the solid medium inoculated with a testing microorganism and incubating it; and calculating the antimicrobial activity according to equation 1 described as in the description.
