Determination of parameters of substances in media comprises illuminating the medium, spectrally resolving the reflected and backscattered light, and approximating the amount of reflected and backscattered light by a model function that includes both the amount of light backscattered by the medium without fully penetrating the medium and the amount of light reflected by a background and fully transmitted through the medium. An Independent claim is also included for an arrangement for performing the determination, comprising an illuminated slit and an optical system, including a pivotable mirror, that creates an image in the form of a series of strips behind the eye and confocally transmits the light reflected or backscattered by the strips to the entrance slit of a polychromator, the output from which, corresponding to sequentially illuminated and scanned strips, is connected via a detector to a computer.
