PROBLEM TO BE SOLVED: To eliminate trouble in focusing at the time of observation by providing a holding member for holding cylindrical culture vessels at prescribed intervals on a stage for placing a sample. SOLUTION: A holding part 60 is provided with a stage attaching member 61 and a roller (holding member) 62 and is respectively provided on both the left and right surfaces of a stage 31 confronted with an optical axis through an objective lens 32 inbetween. The stage attaching member 61 is composed of orthogonal side plate 61a and bottom plate 61b and rollers 62 are provided at prescribed intervals in the lengthwise direction of the side plate 61a. This interval is determined from the diameter of a cylindrical culture vessel 50 held by the roller 62. The roller 62 is attached to a shaft 63, which is screwed on the side plate 61a, so as to be rotated and a washer 64 is provided between the roller 62 and the side plate 61a. The stage attaching member 61 is fixed on the stage 31 by a bolt 65 with hexagonal hole screwed through the bottom plate 61b and the cylindrical culture vessel 50 is held by four rollers 62.
