PROBLEM TO BE SOLVED: To provide a chemiluminescence method and a chemiluminescent reagent, both enabling highly precise measurement of a peroxidase to be carried out in a wide range of peroxidase concentration. SOLUTION: When chemiluminescently measuring a peroxidase in a specimen by formulating the specimen containing the peroxidase with a reagent comprising (a) 2,3-dihydro-1,4-phthalazinedione, (b) a chemiluminescence potentiator and (c) an oxidizing agent, this method is characterized by making the ingredients (a) and (b) exist in their concentrations meeting the following formulas (1) and (2) [wherein, X is the concentration (mM/L) of the ingredient (b); and Y is the concentration (mM/L) of the ingredient (a)] in oxidation reaction: (1) 1.0<=X<=2.6 and (2) 1.3X<=Y<=12.0X. This chemiluminescent reagent for measuring a peroxidase is characterized by comprising a reagent solution (the first solution) where ingredients (a) and (b) are dissolved in concentrations meeting the formulas (1) and (2) in oxidation reaction in a buffer solution and an aqueous solution of the ingredient (c) (the second solution) as constituent reagents.
