PROMOTER OF PYRUVATE DECARBOXLASE GENE OF ASPERGILLUS ORYZAE, RECOMBINATION PLASMID AND TRNSFECTED MICROBES

摘要

PURPOSE: Provided are promoter of pyruvate decarboxylase gene of Aspergillus oryzae and a preparation method for transfected microbes by introducing the vector having the promoter into the microbes. Thereby, useful protein of Aspergillus oryzae is mass produced. CONSTITUTION: A transfected microbes are prepared by the following steps of: preparing genomic DNA library of Aspergillus oryzae; selecting genomic DNA library by using pyruvate decarboxlase gene signature as probe which is find in great quantities in gene signature produced from vegetative reproductive cells of Aspergillus oryzae; separating promoter of pyruvate decarboxylase gene by Southern blot hybridization method and detecting sequence thereof; and introducing plasmid, in which specific gene to be expressed is linked to the downstream of the promoter, into a strain by using the promoter of pyruvate carboxylase.
