PROBLEM TO BE SOLVED: To obtain the subject fiber capable of efficiently providing the section having a nucleic acid arbitrarily and accurately arranged in a high density, in good reproducibility, and useful for detection or the like of the kind and the amount of the nucleic acid by allowing a void part of the porous fiber to hold a nucleic acid- immobilized gel. SOLUTION: The objective nacleic acid-immobilized gel-holding porous fiber is obtained by biotinylating a probe aminated by introducing NH2(CH2)6 group to 5' terminus of an oligonucleotide by using biotin amidite, adding the obtained oligonucleotide having the biotin group at the 5' terminus to an aqueous solution containing acrylamide, methylenebisacrylamide, 2,2'-azobis(2-amidinopropane) dihydrochloride and an avidinized agarose, dipping a porous fiber made of a polyethylene or the like in the obtained aqueous solution, transferring the dipped porous fiber into a hermetically sealed container the interior of which is saturated with steam, and allowing the fiber to stand still at 80 deg.C for 4 hr to carry out a polymerization reaction. The nucleic acid-immobilized gel-holding porous fiber holds the nucleic acid- immobilized gel at the void part of the porous fiber, and is useful for detecting the kind and the amount of the nucleic acid in a specimen. |
