PROBLEM TO BE SOLVED: To eliminate impurity contamination in a fixing and supporting agent as much as possible by filtering glucomannan prepared by oxidizing and dissolving refined powder of an arum root with diluted nitric acid, depositing, agglutinating and precipitating glucomannan in alcohol, and collecting it. SOLUTION: Refined powder of an arum root is prepared by milling crude powder of an arum root, from which a black skin part is peeled off thickly, and drying it, and 10g of the refined powder is dissolved in 100-200 ml of water and heated at 50-80 deg.C, and then, put in methanol so as to be agglutinated and precipitated so as to be collected. The collected material is dried and water-soluble impurities other than glucomannan are eliminated to a certain extent. In this process, water-soluble impurities, which are not agglutinated or precipitated and eluted in water, such as water-soluble protein and carbohydrate are eliminated. In fixing and supporting of a biopsy sample by means of various supporting agents made of glucomannan modified derivatives, an embedding cassette 1 is placed on a Petri dish 2, a biopsy sample 3 is stored in the embedding cassette 1, and a supporting agent in a fixing and supporting agent container 4 is poured in the arrow A5 direction onto the biopsy sample 3 in the embedding cassette 1, and consequently, the biopsy sample 3 in the cassette 1 is embedded with the supporting agent 5.
