FUNGUS SYMBIONT OF NEMATODE

PURPOSE: Fungus symbiont of nematode is provided which is used as a biocontrol agent of nematodes causing plant diseases. Thereby plant diseases are prevented. CONSTITUTION: Fungus symbiont, Bursaphelenchus xylophilusis, is isolated from dead nematode and plated on 2% of water agar plate. Using microscope, conidia attached on nematode are separated and transferred to PDA plate. The isolated fungus is cultured at 25°C for 2-3 weeks and examined morphology with microscope. The morphology of sporangium and conidia is examined with microscope. Fungus symbiont is keep on PDA slant at 4°C or room temperature. The fungus is cultured on one twentieth intensity of CMA plate at 25°C for 2-3 weeks and a pieces of agar containing infectious fungi is put on 2% water agar plate inoculated with nematode. The plate is examined with light microscope and infection rate of nematode is checked. The diameter of colony on PDA plate incubated at 25°C for 7 days is 3.5-4.5 mm with ash color. The mycelium is branched and has septation. The fungus is named Esteya vermicola.
