The invention relates to a method of cloning a target nucleic acid sequence by performing two PCR reactions to generate two FCR products differing in terminal sequences. The PCR products thus obtained are denatured and annealed together to form a heteroduplex, which is ready for ligation and transformation of bacteria without ay restriction digestion.
