FORMATION OF SOMATIC CELL EMBRYO BY SUSPENSION CULTURE OF ELEUTHEROCOCCUS CENTICOSUS MAXIM AND MASS PRODUCTION METHOD OF MICROPLANT

PURPOSE: Formation of somatic cell embryo by suspension culture of Eleutherococcus centicosus and a mass production method of microplant are provided. CONSTITUTION: Seed is stored at 10°C for 18 months and the size of embryo reaches about 7mm. The zygote embryo is taken under sterile condition and induced germination on half Murashige and Skoog's medium containing 0.7 % of agar. Cotyledon grown to about 2cm long for 2 weeks is cut and incubated on Murashige and Skoog's medium containing 1mg/ml 2,4-D, 3% of sucrose and 0.7% of agar at pH 5.8. Over 30 somatic cell embryos are produced from one cotyledon. The embryo is incubated on the same medium and callus is formed. The callus is sustained by successive culture of callus every 5 weeks and somatic cell embryo is obtained by incubation on Murashige and Skoog's medium without 2,4-D. Microplant is obtained by incubation on one third Murashige and Skoog's medium containing 1% of sucrose and 0.7% of agar with germinated somatic cell embryo on the Murashige and Skoog's medium containing 1mg/ml GA3, 0.7% of agar and 3% of sucrose.
