PURPOSE: Novel calpaines, their use and a production method thereof are provided. Further a screening method for new calpaine inhibitors and the use thereof are provided. CONSTITUTION: Calpaines belong to non-lysosome of cystein protease group, involve in Ca dependent signal-transference in eukaryote cells, therefore regulate physiologically cellular functions. In high concentration of Ca, calpaines are over-activated and cause pathological problems such as geriatric diseases. Thus, calpaine inhibitors are useful for treating such diseases. A method for preparing calpain inhibitors comprises the following steps of: isolating calpain nCL-3 from tissues or cells expressing enzyme nCL-3; measuring inhibitions of enzyme nCL-3 substrates and one or more enzyme calpain I and/or II substrates; and screening selective calpain inhibitors. nCL-3 calpain gene comprises SEQ ID NO: 1, allele mutants having homology of 60-100% with amino acids derived from SEQ ID NO: 1, homologues, or derivatives
