NOVEL PEPTIBOL ANTIBIOTIC PRODUCED FROM ASCOMYCETES APIOCREA SP. 14T STRAIN, PREPARATION METHOD AND USE THEREOF

PURPOSE: A novel peptibol antibiotic (KGT14-3) produced from Ascomycetes Apiocrea sp. 14T strain (deposit No. KCTC 8921P) is provided which shows an antiviral activity to tobacco mosaic virus, a cytotoxic antitumor activity to the cell of pancreatic tumor and melanoma and an antibacterial activity to the gram-positive bacteria, therefore can be used in an antiviral agent, an antitumor agent and an antibacterial agent. CONSTITUTION: A peptibol antibiotic (KGT14-3) is represented by structural formula (I): AcPhe-Aib-Ala-Aib-Iva-Leu-Gln-Gly-Aib-Aib-Ala-Ala-Aib-Pro-Iva-Aib-Aib-Gln-Trpo l. The preparation method of the peptibol antibiotic KGT14-3 comprises the steps of: culturing the Apiocrea sp. 14T in potato glucose agar plate comprising 0.1 % of lactic acid for 3-7 days at 25-28°C followed by isolating the strain; inoculating each rice medium with the fragment of the strain and then incubating for 20-30 days at 25-28°C; homogenizing and extracting with ethanol; concentrating the extract under vacuum to remove an organic solvent; and purifying the KGT14-3 by performing chromatography such as silica gel column chromatography, thin-layer chromatography, high-pressure liquid chromatography.
