PROBLEM TO BE SOLVED: To obtain a new protein comprising a protein having a specific amino acid sequence, having Na+-HCO3- cotransporter activity and useful for developing a prophylactic and therapeutic drug, a diagnostic drug, or the like, for diseases such as myocardial infarction, unstable angina, hypertension, cerebral infarction, cerebral hemorrhage. SOLUTION: This protein has an amino acid sequence, which is same or practically same as the amino acid sequence represented by the formula, and Na+-HCO3- cotransporter activity. The protein is useful for developing a prophylactic drug and a therapeutic drug, a diagnostic drug, or the like, for diseases such as myocardial infarction, unstable angina, cardiac dysrhythmia, cardiac failure, hypertension, cerebral infarction, cerebral hemorrhage, subarachnoid hemorrhage, or the like, by screening a compound promoting or inhibiting the Na+-HCO3- cotransporter activity. The protein is obtained by screening a plasmid cDNA library originated from a human heart using a synthetic oligonucleotide comprising a partial sequence as a probe, incorporating the obtained gene into a vector, transducing the vector into a host cell such as CHO-K1 cell and expressing the gene. |
