PROBLEM TO BE SOLVED: To determine dietary fibers in various samples more simply and easily than conventional in an improved state in decreasing troublesomeness and processing time by determining dietary fibers using a thermoduric α-amylase, a neutral protease and amyloglucosidase. SOLUTION: When determining dietary fibers using a thermoduric α-amylase, a neutral protease and amyloglucosidase, first of all the thermoduric α-amylase is made to act on a sample under a condition of pH 5.5-6.5, and then the neutral protease and amyloglucosidase are together made to act on the sample under a condition of pH 5.5-6.5 to prepare an enzymatically treated liquid. Then the liquid is added with 95 vol.% ethanol of four times the quantity of the liquid and is left as it is for 1 h. at a room temperature to precipitate dietary fibers. The dietary fibers are washed, purified, dried and weighed to determine the dietary fibers in a sample. |
