PRODUCTION OF GENE RECOMBINANT HUMAN THYROID STIMULATING HORMONE RECEPTOR OR ITS DERIVATIVE USING INSECT CELL

PROBLEM TO BE SOLVED: To stably and in large quantities obtain gene recombinant human thyroid stimulating hormone receptor(TSHR) used in the measurement or the like of an anti-TSHR autoantibody by cultivating an insect cell infected with a virus containing a gene of human TSHR or the like under a high oxygen concentration. SOLUTION: The objective gene recombinant human thyroid stimulating hormone receptor(TSHR) or its derivative useful in the measurement or the like of an anti-TSHR autoantibody for diagnosing thyroid autoimmune diseases represented by Basedow disease is obtained by cultivating an insect cell infected with a virus containing a gene coding for human TSHR or its derivative under a high oxygen concentration condition to produce insoluble human TSHR or its derivative, solubilizing the resultant insoluble human TSHR or its derivative in the presence of a recfucing agent and a protein denaturing agent and not only separating it from impurities but also decreasing the concentration of the above reducing agent and protein denaturing agent.
