PLASMIDS, THEIR CONSTRUCTION AND THEIR USE IN THE MANUFACTURING OF INTERLEUKIN-4 AND INTERLEUKIN -4 MUTEINS

PURPOSE: Expression vectors for interleukin(herein referred to IL)-4 and IL-4 muteins are provided which has high stability and expression rate. Also, a manufacturing method thereof id provided. Recombinant IL-4 and IL-4 muteins are prepared at high yields by using microorganisms transformed by the plasmids. CONSTITUTION: An expression vectors comprise the followings in direction of 5' to 3'. They are as follows: E. coli phage T5 promoter, controllable promoter consisting of two lac operator sequences, ribosome-binging site derived from E. coli phage T7 g10, translation initiation codon, structural gene of IL-4 or IL-4 mutein, and transcription terminator located downstream of the structural gene. The recombinant vectors are highly stable and express IL-4 or IL-4 mutein at an enhanced efficiency. Thus, recombinant IL-4 and IL-4 mutein can be mass-produced by involving the recombinant microorganisms which are transformed with the vectors.
