The recombinant interleukin IL-4 muteins have at least one amino acid substitution in the binding surface of either the A- or C- alpha helices. The amino acid substitutions are in the A -helix are selected from positions 5, 11, 13 and 16, and in the C-helix, positions 77, 78, 81, 84, 85 (except for R85Q) and 89. These muteins bind to the IL-4R alpha receptor with greater affinity than native IL-4 and can be used in the treatment of immune disorders, cancers or tumors, abnormal cell growth or immunomodulation.
|
