PURPOSE: A kit is provided which attaches HCV 3` terminal RNA to a Covalink by a covalent bond so that it measures HCV replicase activity in an exact and massive way. CONSTITUTION: A kit contains RNA with HCV 3` terminal, RNA replicase in HCV, Covalink module, biotin-16-UTP, streptavidin-alkali phosphatase adhesives. The measurement method is as follows: (1) Covalent bond step wherein a RNA with HCV 3` terminal is added to a Covalink module to form a covalent bond between hexane and module; (2) Replication reaction step which a HCV replication enzyme, rNTP, biotin-16-UTP and reaction buffer solution are added to the Covalink module combined by RNA in a HCV 3` terminal to replicate; (3) Coloring reaction step which washes a well and reacts residual biotin-16-UTP to generate a color; and (4) Spectroscopic analysis step which measures the extent of coloring.
