Parathyroid hormone analogues for use as medicaments in treating osteoporosis

Analogues of human parathyroid hormone (hPTH) that have increased activities in bone restoration and increased bioavailabilities are disclosed. The peptides described are derivatives of hPTH-(1-31) that are cyclized, for example, by formation of lactams between either Glu22 and Lys26 or Lys26 and Asp30. In addition, either a Leu or other hydrophobic residues, such as Ile, norleucine, Met, Val, Ala, Trp, or Phe, may substitute the natural Lys27. Using the ovariectomized rat model, these analogues have been shown to have enhanced abilities to stimulate adenylyl cyclase in rat osteosarcoma cells, and show increased activities in bone restoration. The use of hPTH-(1-31) in the manufacture of a medicament for treating osteoporosis is also disclosed.
