The method of evolving a polynucleotide thru DNA shuffling for acquisition of a desired property comprises: a) providing a population of template polynucleotides and a pair of first and second primers capable of hybridizing to at least one of the strands of the template polynucleotides and its complement; b) conducting a multi-cyclic polynucleotide extension reaction of the template polynucleotides using the first primer only under conditions of incomplete extension; the incomplete extension product anneals to different template polynucleotides in different extension cycles and undergoes further extensions; generating recombinant incomplete extension products and subsequently full-length recombinant extension products; c) contacting the full-length recombinant extension products with the second primer and extending the second primer to form a double stranded full-length recombinant extension products.
