PROCESS FOR MANUFACTURING RUMBROKINASE LIQUID AND USE THEREOF

PURPOSE: A manufacturing method of lumbrokinase that can be used for thrombus dissolving agent is provided, which decreases bleeding complication of existing thrombus dissolving agent, by which one can get pure lumbrokinase for medicinal use. CONSTITUTION: The manufacturing method of lumbrokinase comprises the steps of: mixing Lumbricus Rubellus dried powder and saline or Tris(pH 8.0) to synthesize protein at 4-30deg.C by shaking with a magnetic stick; centrifuging the synthesized protein for 30minutes to remove impurities; adding ammonium sulfate to bind with the protein, and removing the unnecessary protein by centrifuging; removing extra ammonium sulfate by dialysis membrane; refining the protein having fiber-disintegration ability from the first stage protein by DEAE-cellulose anion-exchange chromatography; washing, and gathering the protein on the column; and refining the pure lumbrokinase from the second stage protein by para-amino benzamidine cepharose 6B affinity column chromatography.
