PROBLEM TO BE SOLVED: To quantitatively measure a trace amount of protein in a liquid sample such as urine. SOLUTION: This composition contains indium or an indium compound, a pigment capable of forming a complex with indium, and a buffering agent retaining the pH of a system to acidic pH. Here, a sufficient color variation for indicating the existence and/or a concentration of a protein in the sample is generated by contacting the composition with a protein-containing liquid sample. The pigment capable of forming the complex with indium preferably includes a polyhydroxybenzenesulfon phthalein type pigment and/or a polyhydroxybenzene phthalein type pigment.
