The present invention relates to methods and compositions for modulating the release of cytokines by alpha beta 7-expressing cells. Binding polypeptides and other molecules which selectively bind to alpha beta 7 and, thereby, block or enhance the Th2 cytokine response by the alpha beta 7-expressing cell, also are provided. The antagonists and agonists of the invention are useful in screening assays to identify pharmaceutical lead compounds which are capable of modulating cytokine release by alpha beta 7-expressing cells. Methods and pharmaceutical compositions for modifying the Th2 cytokine response by an alpha beta 7-expressing cell, in vitro and in vivo also are provided.
