MEASUREMENT OF ENZYME ACTIVITY BY UTILIZING CHANGE OF PERMEABILITY

PROBLEM TO BE SOLVED: To measure the activities of an endo-type hydrolytic enzyme in an enzyme solution by adding exo-type and endo-type hydrolytic enzymes of a specific substrate to a gel containing the dispersed substrate and having a turbidity, and measuring the decreasing rate of the turbidity. SOLUTION: An enzyme solution containing an exo-type hydrolytic enzyme (e.g. exo-type amylase) and an endo-type hydrolytic enzyme (e.g.α-amylase), capable of degrading a specific substrate (e.g. starch) is added to a gel (e.g. agar) prepared by allowing the substrate to be dispersed to have a turbidity, and the decreasing rate of the turbidity caused by the addition of the enzyme solution is measured to rapidly and readily measure the activities of the endo- type hydrolytic enzyme in the coexistence of the exo-type hydrolytic enzyme.
