PROBLEM TO BE SOLVED: To obtain N-formyl leucine used for peptide synthesis at a high purity by specifying the final pH of a reaction medium after the reaction is completed. SOLUTION: In a reaction of leucine (A) with formaldehyde (B), after the reaction is completed, the reaction medium is mixed with water and an acid (e.g. hydrochloric acid) and the final pH of the mixture is adjusted to about 2-3. The reaction of the component A with the component B is performed using <4 mol component B based on 1 mol component A in the presence of about 0.1-12 mol formic acid based on 1 mol component A. The amount of water used for forming precipitate is about 1.5-5 pts. mass based on the mass of the component B. Preferably at the time of forming the precipitate, water is firstly mixed with the reaction medium and subsequently the pH of the mixture is adjusted to about 2-3 by adding the acid and maintained in the same pH range.
