<p>Methods and materials for filling perforations in partially demineralized cortical bone allografts with matrices including a biodegradable, biocompatible polymer, preferably in conjunction with osteoconductive, filler or bulking agent and/or buffering material, such as HA, are disclosed. The matrix enhances control of resorption and bony ingrowth in the cortical bone allografts. The matrix can be used to deliver factors to bone progenitor cells in vivo to stimulate bone. The matrix can also be used to transfer the stimulatory factor into the cells of the soft tissues surrounding the cortical bone allograft or cultured cells or recombinant cells maintained in vitro, where all that is required is to add the stimulatory factor composition to the cells, e.g., by adding it to the culture media. The matrix may be employed to promote expression of a desired gene in bone cells or tissues surrounding the cortical bone allograft and to impart a particular desired phenotype to the cells.</p> |
