PROBLEM TO BE SOLVED: To obtain a plasmid which can be amplified in a rumen microorganism and can be used as a shuttle vector between Streptococcus bovis and Escherichia coli by incorporating a self-replicative sequence derived from S. bovis. SOLUTION: A plasmid is provided which has the restriction enzyme map shown by formula I, has a size of 3904 bp, contains a self-replicative sequence derived from a bacterium Streptococcus bovis which degrades many kinds of soluble saccharides, starch, pectin, protein, and so on in the rumen, contains (A) DNA consisting of the base sequence shown by formula II or (B) DNA which can hybridize to DNA consisting of the base sequence shown by formula II under the stringent condition, and is self-replicative. Escherichia coli containing the plasmid were deposited as FERM P-17008 and 17009. |
