PROBLEM TO BE SOLVED: To provide a safe phospholipase C, capable of being advantageously applied to food and having good pH stability and good thermal stability. SOLUTION: This phospholipase C is produced from a mold comprising Aspergillus niger AC782 and has the following enzymochemical properties. (1) Action: hydrolyzes the 3-phosphate ester group of a phospholipid to release and produce a diacylglycerol and a phosphoryl salt group. (2) Substrate characteristics: when the hydrolysis activity of the phospholipase C for phosphatidyl choline is 100, the relative activity of the phospholipase C is 200 for phosphatidyl ethanolamine, and 120 for phosphatidic acid. (3) The optimal pH: 3.3-3.7. (4) The optimal temperature: 53-57 deg.C. (5) The pH stability: pH 2.0-5.0. (6) Thermal stability: 40-60 deg.C. (7) Mol.Wt.: about 54,000. (8) Isoelectric point: p14.3±0.1 (9) Michaelis constant Km: 5.6 mM (measured for the phosphatidyl choline substrate at pH of 4.5 at 37 deg.C). |
