PURPOSE:To produce the subject enzyme having a specific activity on alpha-1,2- mannoside bond and useful for diagnosis, therapy, etc., of mannosidosis by culturing an alpha-1,2-mannosidase-producing bacterium belonging to Bacillus genus and collecting the products from the cultured material. CONSTITUTION:A bacterium (e.g. bacillus sp. M-90 strain) belonging to Bacillus genus and having alpha-1,2-mannosidase productivity is cultured and alpha-1,2- mannosidase is collected from the cultured material to obtain the objective enzyme capable of specifically hydrolyzing alpha-1,2-mannoside bond at non-reducing end positions of the saccharide and liberating mannose in the case of using alpha-mannane or an oligosaccharide containing alpha-1,2-mannoside bonds as the substrate, capable of synthesizing an oligosaccharide containing alpha-1,2-mannoside bonds by dehydration condensation of mannose in the case of using mannose as the substrate, having 5.5-7.0 optimum pH, 5.0-8.0 stable pH range, 40-50 deg.C optimum temperature, <=40 deg.C stable temperature range, receiving an inhibitory effect by EDTA or Hg2+ and capable of activation by Ca<++>. |
