PURPOSE: A manufacturing method of 6-O-methylerythromycin A form II is provided which enables to obtain highly pure 6-O-methylerythromycin A form II in high yields. CONSTITUTION: 6-O-methylerythromycin A form II can be obtained by the recrystallizing of 6-O-methylerythromycin A(formula 1). Recrystallizing method comprises the following steps. They are as follows: a) 6-O-methylerythromycin A is solubilized by using an acid aqueous solution or the solvent mixture of an acid aqueous solution and an organic aqueous solution; b) alkali is added into the solution to form precipitation; c) the precipitates are vacuum dried at the temperature of 45-50°C. The acid can be selected from inorganic acid such as hydrochloride, sulfuric acid or nitric acid, or organic acid such as acetic acid or formic acid, and the organic aqueous solution can be C1-C5 alkanol such as methanol, ethanol, n-propanol, isopropanol, n-butanol, isobutanol or t-butanol, tetrahydrofuran or acetonitrile. The alkali added into the solution of 6-O-methylerythromycin A is sodium hydroxide.
