PURPOSE: A bacterium which has an ability to produce an amino acid and in which a novel gene (rhtB) coding for a protein having an activity of making a bacterium having the protein L-homoserine-resistant is enhanced, is cultivated in a culture medium to produce and accumulate the L-amino acids of E.coli, specially L-homoserine, L-alanine, L-isoleucine, L-valine, and L-threonine in the medium, and the amino acid is recovered from the medium. CONSTITUTION: 86min region of E.coli chromosome is cloned by a multi copy vector such as plasmid vector(pBR322, pMW118 or pUC19) or phage vector (lambda 1059, lambda BF101, M13mp9) to provide E.coli with L-homoserine resistance and E.coli having L-homoserine resistance to the high concentration of L-homoserine by amplifying the region can produce L-amino acids in high yields and accumulate them in the media. A bacteria strain belong to Escherichia sp. which produces target amino acid is used for produce target amino acid by increasing Rh(Protein related with L-homoserine resistance) activity.
