PROBLEM TO BE SOLVED: To produce a poly-3-hydroxyalkanoic acid usable for alternative plastics by introducing a polymerase gene of the poly-3-hydroxyalkanoic acid, and a fatty acid synthetase gene to a host microorganism, proliferating the host microorganism, and isolating and purifying the product. SOLUTION: A polymerase gene of a poly-3-hydroxyalkanoic acid is incorporated into a wide host range expression vector pJRDTrc1 for a Gram-negative bacterium to construct the expression vector of the polymerase gene of the poly-3-hydroxyalkanoic, acid. On the other hand, a fatty acid synthetase gene derived from Escherichia coli is incorporated into a vector pBluescriptII-KS(+) for expression, to construct an expression vector of the fatty acid synthetase gene. These expression vectors are transduced to a host microorganism such as Escherichia coli to transform the microorganism, and the transformed microorganism is proliferated in the presence of a carbon source. The product is isolated from the proliferated microorganisms and purified to provide the objective poly-3-hydroxyalkanoic acid useful as alternative plastics. By this method, highly pure poly-3-hydroxyalkanoic acid is readily and cheaply produced in large scale. |
