PURPOSE: A method for producing hepatitis B virus surface antigen(HBsAg) proteins is provided, therefore the hepatitis B virus vaccine can be developed using HBsAg to improve problems in conventional hepatitis B virus vaccines. CONSTITUTION: The method for producing hepatitis B virus surface antigen(HBsAg) comprises the steps of: preparing a HBsAg expression vector CMV4U for mammalian cells by inserting the gene encoding preS1, preS2 and S regions of HBsAg between CMV promoter and bovine growth hormone polyadenylation gene and further inserting Neo gene and dihydrofolate reductase(DHFR) gene; transforming Chinese Hamster Ovary(CHO) with CMV4U to produce CHO cell line 1B3(KCTC 0416BP); and incubating CHO cell line 1B3(KCTC 0416BP) and purifying the supernatant of the fermented medium to obtain HBsAg protein containing preS1, preS2 and S regions.
