PURPOSE: Provided is a method for purifying rabbit prothrombin kringle-2 obtained from rabbit's serum. The rabbit prothrombin kringle-2 can prevent endothelial cells from growing and is useful in treating rheumatoid arthritis and cancer. CONSTITUTION: The method for purifying rabbit prothrombin kringle-2 is comprised of the next steps of: i) obtaining serum containing rabbit prothrombin kringle-2 by injecting Bacillus Calmette-guerin into a New Zealand white rabbit; ii) precipitating the serum in 50-90% ammonium sulfate and carrying out DEAE-Sepacell ion exchange chromatography; and iii) subjecting segments that block the growth of endothelial cells to Sepacell S-200 gel filtering chromatography, FPLC mono-Q ion exchange chromatography, CM-cibacron Blue 3GA chromatography, and FPLC Superose 12H/R gel filtering chromatography. The rabbit prothrombin kringle-2 has 22,000Da and 12,600Da of molecular weight measured by SDS-PAGE and mass spectrograph, respectively. Its isoelectric point is pH5.0.
