METHOD FOR PURIFICATION OF HEPATITIS B VIRUS SURFACE ANTIGEN

PURPOSE: Provided is a purification method of the recombinant hepatitis B surface antigen, thereby hepatitis B surface antigen is purified with high purity. CONSTITUTION: A purification method of a recombinant hepatitis B surface antigen is composed of the following steps of: (a) solubilizing yeast cell wherein hepatitis B surface antigen is expressed under surfactant and then supernatant containing hepatitis B surface antigen is separated; (b) contacting the resultant with a silica and then detaching hepatitis B surface antigen by contacting the silica with buffer solution of pH 8.8 to 11.0; (c) passing the resultant solution through anion-exchange resin equilibrated with buffer solution that contains oxide then followed by washing it with buffer solution that contains NaCl of 0.05 to 0.1 M and eluting hepatitis B surface antigen with buffer solution that contains NaCl of 0.15 to 0.5 M; and (d) performing gel permeable chromatography with fraction that contains hepatitis B surface antigen.
