FUSION PROTEIN PRES2HV OF SURFACE ANTIGEN PRES2 SAG OF HEPATITIS B VIRUS AND HV-H5 PROTEIN OF HEPATITIS C VIRUS, AND PRODUCING METHOD THEREOF

PURPOSE: A fusion protein preS2HV of the surface antigen preS2 SAg of hepatitis B virus and HV-H5 protein of hepatitis C virus and a producing method thereof are provided, thereby vaccines having improved specificity and immunity for HBV and HCV can be produced. CONSTITUTION: The fusion protein preS2HV represented by the amino acid sequence of Fig. 1 is produced by the steps of: preparing a primer for pre S2 SAg gene of HBV; amplifying the pre S2 SAg gene by PCR(polymerase chain reaction) using plasmid pYPSAG100 as a template; coupling the amplified pre S2 SAg gene with the ubiquitin gene and amplifying the coupled gene by PCR; cleaving the coupled gene with endonucleases and inserting the fragments into an appropriate expression vector; preparing a primer for HV-H5 gene of HCV; amplifying the HV-H5 gene by PCR using plasmid M13mp18-LBC-HV(ATCC 75448) as a template; and inserting the amplified HV-H5 gene into the expression vector containing the coupled gene of pre S2 SAg and ubiquitin.
