PURPOSE: Provided is DO-stat fed batch culture method for manufacturing hPTH (human parathyroid hormone) in a high yield but at a low cost by changing a medium along with the addition of L-arabinose. CONSTITUTION: DO-stat fed batch culture method for manufacturing hPGH (human parahyroid hormone) is comprised of the next steps of: i) cultivating E. coli MC1061:pm153PTH (KCCM-10102) in a culture medium at 30 deg.C for 15 hours at 150rpm and then, in a fermenter at 37 deg.C and pH 7 at the speed of 600rpm; and ii) cultivating the E. coli through DO-stat fed batch culture when DO (dissolved oxygen) starts increasing sharply while adding L-arabinose. In particular, only when DO reaches over 50%, composition of fed batch culture is added. And the E. coli is transformed by an expression vector having a fusion gene of phosphoribulokinase (PRK) and human parathyroid hormone (hPTH).
