PROBLEM TO BE SOLVED: To quickly detect the fluctuation of the salt-tolerance ammonia oxidative bacterium in active sludge by utilizing the antigen-antibody reaction of an antibody peculiar to the salt-tolerance ammonia oxidative bacterium. SOLUTION: New bacterium belonging to Nitrosomonas, for example, and capable of oxidizing ammonia efficiently and stably even at a high salt concentration are manufactured as an antigen. This antigen is cultured with a rabbit or a goat used as an immune animal, for example, exsanguination is made to obtain an antisera when the titer of an antibody is fully increased, and a proper refining process is applied to it to generate the antibody having a sufficient peculiarity to salt-tolerance ammonia oxidative bacterium. The active sludge or processed water in an object device are collected and unified, then it is diluted with purified water or a physiological salt solution in several stages to obtain a measurement sample. The generated antibody is used against this measurement sample to quantitify the salt-tolerance ammonia oxidative bacterium via the peculiar-antibody reaction. |
