PROBLEM TO BE SOLVED: To obtain a novel chimera antibody against human interleukin-6 receptor that comprises a chimera L-chain having the L-chain C region of human antibody as well as the L-chain V-region of mouse monoclonal antibody against human interleukin-6 receptor, thus shows low immunogenicity and is useful as a therapeutic agent for myeloma. SOLUTION: This chimera antibody has a novel chimera L-chain that has the L-chain C-region of human antibody and the L-chain V-region of mouse monoclonal antibody against human interleukin-6 (IL-6) receptor, where this L-chain V-region has amino acid sequences of formula I to formula III. When it is dosed to human, the chimera antibody shows low immunogenicity and is useful as a therapeutic agent or the like for myeloma. The chimera antibody is prepared by linking a cDNA clone encoding the L-chain and H-chain V-region of mouse monoclonal antibody against the human IL-6 receptor to a cDNA encoding the human C region connecting to the expression vector for mammalian cells to prepare a recombinant expression vector, transducing the vector into a mammalian cell, for example, COS cell or the like and expressing the vector. |
