PURPOSE: Provided are new polyketides, especially, erythromycins having anti-bacterial, anti-protozoal activities and a method for producing these compounds using genetic engineering techniques. CONSTITUTION: Novel erythromycin derivatives have been substituted with R1 at C-13 site on the microscope ring; R1 = alkyl, alkenyl, iso-alkyl, cyclic groups such as C3-C6 cycloalkyl or cycloalkenyl groups, which contain one or more substitutes with hydroxyl, or C1-4 alkyl, alkoxy, or halogen groups. A productions of specific erythromycin analogs are accomplished by growing a culture of a polyketide-producing microorganism, Saccharopolyspora erythraea, which has been manipulated to have the vectors expressing genetically mutated genes involved in erythromycin biosynthesis, with R1COOH-containing medium in anaerobic condition, and by isolating the specific erythromycin analogs from the cell culture. |
