PROBLEM TO BE SOLVED: To dye melanin easily and surely by dyeing melanin using basic silver nitrate solution of specific concentration. SOLUTION: The concentration of basic silver nitrate solution is set to approx. 0.1-1 w/v% and its pH is adjusted to approx. 10-10.5 with a base such as aqueous ammonia and the like. In conducting observation with particular emphasis on the localization of large melanin, the concentration of silver nitrate is set to low concentration of approx. 0.1 w/v%. On the other hand, in conducting observation with particular emphasis on the distribution of fine melanin, it is set to high concentration of approx. 1 w/v%. Silver-dyeing generally has a large influence of oxidation due to air, however, since a low concentration of the silver nitrate is used, so it permits the excessive silver nitrate to be washed off rapidly, the influence of oxidation is reduced, and the consumption of the silver nitrate can be reduced. Moreover, the low concentration permits clear dyeing to be attained even at high temperature, dyeing time is reduced to approx. 10 min (about 45 deg.C), excessive dyeing of the melanin can be prevented, and dyeing of only large melamine closely related with flesh color is made possible.
