PROBLEM TO BE SOLVED: To obtain the subject adenosine(uridine)-5',5'''-P1,P4-tetraphosphate that is useful as a starting material for medicines by using adenosine 5'- triphosphate and uridine 5'-triphosphate and isoleucine as reaction substrates and allow isoleucyl-t-RNA synthase to react with them. SOLUTION: Adenosine 5'-triphosphate, uridine 5'-triphosphate and isoleucine are used as reaction substrates and isoleucyl-t-RNA synthase is used as a catalyst in the presence of magnesium chloride in an imidazole buffer solution (pH 7.0) at 40 deg.C for 30 hours to obtain the objective adenosine (uridine)-5',5'''-P1, P4-tetraphosphate (Ap4U) of high concentration in high yield with an industrial advantage. This compound is useful as a medicine or its starting material or the like.
