PURPOSE: A yafB gene that is a gene for 2-keto-D-gluconate producing 2,5-diketo-D-gluconate reductase is defected to prevent metabolism of 2,5-diketo-D-gluconate produced as an intermediate product in the production of 2-keto-L-glonate by transformation in E. coli, thereby production yield of 2-keto-L-glonate as a precursor of vitamin C is enhanced. CONSTITUTION: The yafB gene codes the 2,5-diketo-gluconate reductase gene for production of 2-keto-D-gluconate, which is originated from E. coli. The yafB gene is inserted into a recombinant plasmid pYAFB. The recombinant plasmid pYAFB with the yafB gene inserted thereinto is transformed in a coliform DH5alpha/pYAFB (KCTC 0509BP). In the production of 2-keto-L-glonate as a precursor of vitamin C from gluconate by glucose metabolism, the 2,5-diketo-D-gluconate reductive defective gene is introduced to a host cell to express a glucose reductive in a membrane-bound recombinant strain.
